Allelic diversity in populations of Solanum lycocarpum A. St.-Hil (Solanaceae) in a protected area and a disturbed environment Acta Botanica
Moura,Tânia Maria de; Sebbenn,Alexandre Magno; Martins,Karina; Moreno,Maria Andreia; Oliveira,Giancarlo Conde Xavier; Chaves,Lázaro José; Kageyama,Paulo Yoshio.
This study aimed to compare the genetic diversity of populations of Solanum lycocarpum A.St.-Hil between natural and human disturbed environments, with the assumption that protected areas have greater genetic diversity than disturbed areas. For this study, two populations were sampled in Goiás State, Brazil. One was located in a conservation unit, Serra de Caldas Novas State Park, in the Caldas Novas municipality. The other was located in a pasture area in the municipality of Morrinhos. The two populations are 41 km apart. We sampled 60 individuals from each population, which were genotyped with five microsatellite loci (SSR). The highest number of alleles was recorded in the population of the conservation unit, where we found 11 exclusive and five rare...

Allozymic variation and differentiation in the chilean blue mussel, Mytilus chilensis, along its natural distribution Genet. Mol. Biol.
Toro,Jorge E.; Castro,Grace C.; Ojeda,Johana A.; Vergara,Ana M..
Genetic differentiation in the Chilean blue mussel Mytilus chilensis (Hupé 1854) was investigated based on the variation in the allozyme frequencies of Pgm, Gpi, Icd, Me, Gsr, Lap and Pep in eight samples collected along 1800 km from Arauco (VIII Region) to Punta Arenas (XII Region). Despite the large geographic separations, values of Neis unbiased genetic distance, D (0.004-0.048) and standardised genetic variation among populations, Fst (0.011-0.055) were small. The levels of gene flow (Nm = 8) found in this study prevent the effect of differentiation among populations by genetic drift. This findings indicate that its long-lived planktotrophic larvae provides this species with considerable dispersal ability throughout its range which is favoured by the...

An update on genetically modified crops Ciencia e Investigación Agraria
Schwember,Andrés R.
A. R. Schwember. 2008. An update on genetically modified crops. Cien. Inv. Agr. 35(3):231-250. Genetically modified (GM) crops were introduced in the mid 1990s and two principal transgenic technologies currently domínate the market, herbicide-tolerant (HT) crops and insect-resistant crops (Bacillus thuringiensis (Bt) crops). HT crops have simplified weed management practices, reduced crop production costs, and have had positive effects on the environment. However, there are concerns about the potential development of weeds resistant to glyphosate, the main herbicide employed with HT crops. A second major worry associated with the use of HT crops is the potential introgression of genes from GM crops into wild relatives (i.e. gene flow) and its potential...

Beyond Roadkill, Radiotracking, Recapture and FST—a Review of Some Genetic Methods to Improve Understanding of the Influence of Roads on Wildlife Ecology and Society

Habitat fragmentation continues to occur despite increasing evidence of its adverse effects on ecosystems. One of the major detrimental effects of roads and traffic is the creation of barriers or filters to the movement of wildlife, ultimately disconnecting some populations. Our understanding of the extent to which roads reduce the movement of biota is mostly based on field-based observational methods of inferring animal movement, and to a much smaller extent, on allele frequency-based genetic analyses. Field-based methods, as it is typically feasible to apply them, tend to be informative at fine temporal and spatial scales. Allele frequency-based genetic methods are informative at broad geographic scales but at timescales usually greater than recent...

Biosystematics of the genus Dahlstedtia Malme (Leguminosae, Papilionoideae, Millettieae) Rev. Bras. Bot.
Teixeira,Simone P.; Ranga,Neusa T..
Dahlstedtia has been considered a monotypic genus by some authors, but comprising two species according to others. The latter view was supported by our biosystematic study. The two species are Neotropical, mainly occurring in the Brazilian Atlantic Forest, where they are, however, allopatric. Experimental pollinations show that the two species are intersterile. After interspecificcrossings, few pollen tubes reach the ovule, a dense secretion fills the micropyle and no proembryos are formed. Probably, there is no gene flow between species, but if it should occur, no hybrids would be formed. The present work confirms Burkart's taxonomic view, according to which Dahlstedtia comprises at least two species: D. pinnata and D. pentaphylla.

Combining population genomics and forward simulations to investigate stocking impacts: A case study of Muskellunge ( Esox masquinongy ) from the St. Lawrence River basin ArchiMer
Rougemont, Quentin; Carrier, Anne; Le Luyer, Jeremy; Ferchaud, Anne-laure; Farrell, John M.; Hatin, Daniel; Brodeur, Philippe; Bernatchez, Louis.
Understanding the genetic and evolutionary impacts of stocking on wild fish populations has long been of interest as negative consequences such as reduced fitness and loss of genetic diversity are commonly reported outcomes. In an attempt to sustain a fishery, managers implemented nearly five decades of extensive stocking of over a million Muskellunge (Esox masquinongy), a native species in the Lower St. Lawrence River (Québec, Canada). We investigated the effect of this stocking on population genetic structure and allelic diversity in the St. Lawrence River in addition to tributaries and several stocked inland lakes. Using Genotype‐By‐Sequencing (GBS), we genotyped 643 individuals representing 22 locations and combined this information with forward...

Development of microsatellite loci and population genetics in the bumblebee catfish species Pseudopimelodus atricaudus and Pseudopimelodus magnus (Siluriformes: Pseudopimelodidae) Neotropical Ichthyology
Rangel-Medrano,José David; Márquez,Edna Judith.
ABSTRACT The Neotropical freshwater catfish Pseudopimelodus atricaudus and Pseudopimelodus magnus are two recently discovered species endemic to the Colombian Magdalena-Cauca River basin. In this study, a set of 13 microsatellite loci were developed by using next generation sequence technology to assess the genetic diversity and population structure in P. atricaudus and test for cross-species amplification in P. magnus. Both species exhibited high genetic diversity (P. atricaudus: Na: 9.000 - 9.769 alleles/locus, Ho: 0.760 - 0.804, HE: 0.804 - 0.840; P. magnus: Na: 12.8 - 5.4 alleles/locus, Ho: 0.638 - 0.683, HE: 0.747 - 0.755) compared to the mean levels of genetic diversity reported for Neotropical Siluriformes, and lack of genetic differentiation among...

Development of microsatellite loci and population genetics of the catfish Pimelodus yuma (Siluriformes: Pimelodidae) Neotropical Ichthyology
Joya,Cristhian Danilo; Landínez-García,Ricardo Marcel; Márquez,Edna Judith.
ABSTRACT Pimelodus yuma (formerly Pimelodus blochii) is a freshwater fish, endemic to the Colombian Magdalena-Cauca and Caribbean basins that experiences habitat disturbances resulting from anthropogenic activities. Due to the lack of information about the population genetics of this species, this study developed 14 species-specific microsatellite loci to assess the genetic diversity and population structure of samples from the lower section of the Cauca River. The studied species showed genetic diversity levels higher than the average values reported for Neotropical Siluriformes and significant inbreeding levels as was described for some congeners. Furthermore, P. yuma comprises two coexisting genetic groups that exhibit gene flow along the lower section...

Diversity and genetic structure in natural populations of Hancornia speciosa var. speciosa Gomes in northeastern Brazil Rev. Bras. Frutic.
Martins,Georgia Vilela; Martins,Luiza Suely Semen; Veasey,Elizabeth Ann; Lederman,Ildo Eliezer; Silva,Edson Ferreira da.
Hancornia speciosa Gomes is a fruit tree native from Brazil that belongs to Apocinaceae family, and is popularly known as Mangabeira. Its fruits are widely consumed raw or processed as fruit jam, juices and ice creams, which have made it a target of intense exploitation. The extractive activities and intense human activity on the environment of natural occurrence of H. speciosa has caused genetic erosion in the species and little is known about the ecology or genetic structure of natural populations. The objective of this research was the evaluation of the genetic diversity and genetic structure of H. speciosa var. speciosa. The genetic variability was assessed using 11 allozyme loci with a sample of 164 individuals distributed in six natural populations...

Estimating dispersal and gene flow in the neotropical freshwater turtle Hydromedusa maximiliani (Chelidae) by combining ecological and genetic methods Genet. Mol. Biol.
Souza,Franco L.; Cunha,Anderson F.; Oliveira,Marcos A.; Pereira,Gonçalo A.G.; Reis,Sérgio F. dos.
Hydromedusa maximiliani is a vulnerable neotropical freshwater turtle endemic to mountainous regions of the Atlantic rainforest in southeastern Brazil. Random amplified polymorphic DNA (RAPD) was used to estimate the gene flow and dispersal for individuals inhabiting rivers and streams within a drainage. Nine primers generated 27 scoreable bands, of which 9 (33%) were polymorphic and produced 12 RAPD phenotypes. The gene flow estimates (Nm) among turtles inhabiting different rivers and streams were variable, ranging from 0.09 to 3.00 (mean: 0.60). For some loci, the rates of gene flow could offset population differentiation (Nm > 1), whereas for others random genetic drift could result in population divergence (Nm < 1). Since the genetic variation of...

Evidence of genetic differentiation and karyotype evolution of the sedges Cyperus ligularis L. and C. odoratus L. (Cyperaceae) Acta Botanica
Cruz,Geyner Alves dos Santos; Vasconcelos,Santelmo; Pinangé,Diego Sotero de Barros; Gonçalves-Oliveira,Rodrigo Cesar; Alves,Marccus Vinícius; Torres,Rodrigo Augusto; Benko-Iseppon,Ana Maria.
ABSTRACT The taxonomy of Cyperaceae is complex, with genera like Cyperus harboring species complexes. We analyzed the genetic similarity between Cyperus ligularis L. and C. odoratus L. based on DNA fingerprinting and cytogenetics. Significative genetic differentiation (G ST = 0.363) and low gene flow (N m = 0.877) indicated a clear genetic distinction between the two species. Moreover, the clustering analysis showed two distinct genetic groups, suggesting a lack of evidence for hybridization. The phenogram revealed two different lineages, and although all individuals of C. odoratus were collected from plots close to each other, they possessed greater genetic diversity than that observed among individuals of C. ligularis, which were sampled over a wider...

Evidence of two lineages of the dengue vector Aedes aegypti in the Brazilian Amazon, based on mitochondrial DNA ND4 gene sequences Genet. Mol. Biol.
Lima Júnior,Raimundo Sousa; Scarpassa,Vera Margarete.
Genetic variation was estimated in ten samples populations of Aedes aegypti from the Brazilian Amazon, by using a 380 bp fragment of the mitochocondrial NADH dehydrogenase subunit 4 (ND4) gene. A total of 123 individuals were analyzed, whereby 13 haplotypes were found. Mean genetic diversity was slightly high (h = 0.666 ± 0.029; π = 0.0115 ± 0.0010). Two AMOVA analyses indicated that most of the variation (~70%-72%) occurred within populations. The variation found among and between populations within the groups disclosed lower, but even so, highly significant values. F ST values were not significant in most of the comparisons, except for the samples from Pacaraima and Rio Branco. The isolation by distance (IBD) model was not significant (r = 0.2880; p =...

Frequency of CCR5delta32 in Brazilian populations BJMBR
Vargas,A.E.; Marrero,A.R.; Salzano,F.M.; Bortolini,M.C.; Chies,J.A.B..
A sample of 103 randomly chosen healthy individuals from Alegrete, RS, Brazil, was tested for the CCR5delta32 allele, which is known to influence susceptibility to HIV-1 infection. The CCR5delta32 allele was identified by PCR amplification using specific primers flanking the region of deletion, followed by electrophoresis on a 3% agarose gel. The data obtained were compared to those reported for other populations and interpreted in terms of Brazilian history. The individuals studied came from a highly admixed population. Most of them were identified as white (N = 59), while blacks and browns (mulattoes) were N = 13 and N = 31, respectively. The observed frequencies, considering the white, black and brown samples (6.8, 3.8, and 6.4%, respectively), suggest...
